Mechanism of Action

Tirzepatide is a 39-amino acid synthetic peptide engineered from the GIP sequence with a C20 fatty di-acid moiety that binds albumin and extends its half-life. In receptor-expressing cell lines it acts as an agonist at both the glucose-dependent insulinotropic polypeptide (GIP) receptor and the glucagon-like peptide-1 (GLP-1) receptor, with reported bias toward cAMP signaling over β-arrestin recruitment at GLP-1R. In diet-induced obese rodent models this dual agonism has been associated with improved glucose tolerance, insulin secretion, and energy-balance endpoints beyond single-receptor GLP-1 agonism. Reconstitution & Storage

Lyophilized tirzepatide is stable at −20°C or colder in its sealed vial; −80°C is preferable for long-term archival. Reconstitute in bacteriostatic water (0.9% benzyl alcohol) to a laboratory working concentration of 0.5–2 mg/mL for in vitro use.
